My vote goes to The Jerk, one of the funniest movies ever made.

Pennies From Heaven

It doesn't get much respect, at least that I know of, but Martin is as excellent a dramatic performer (and romantic singer) as he was a comedian. I prefer him in that mold.

Planes, Trains and Automobiles would be a close runner-up.

I'm not voting until I see 'Pennies From Heaven', which I predict will become my favorite film of his.

Of the ones that I have seen, I'm going with 'The Jerk', though I also love 'Planes, Trains, & Automobiles' and 'Shopgirl' (that's right)

Now, I haven't seen The Jerk yet, thats on my Netflix, but out of all the others I gotta go with Bowfinger just because, while this is a Steve Martin question, Eddie Murphy as Kit Ramsey is one of my favorite characters ever, I absolutely cracked up at almost anything he said.

Kit's Agent: This is a great script! Look, it's not Shakespeare, but it...
Kit: Hey, what did you just say?
Agent: I said, 'it's not Shakespeare'...
Kit: 'It's not Shake... ', 'It's not Shake... ' (to Freddy) Do you hear what he's doing?
Freddy: I know he's doing something, I just can't put my finger on it. Yeah, yeah, yeah... What's he doing?
Kit: Shakespeare, Freddy, Shakespeare!
Freddy: Shakespeare?
Kit: Shake a spear! Spearchucker! I'm a spearchucker now!

Hard to pick because there have been so many great ones.

Planes, Trains & Automobiles edges The Jerk by a hair in my book.

I have a soft spot for Roxanne - I just love the "Twenty somethings better" scene where he comes out with insults for the "Dick." Love that scene.

I think the Jerk is probably the best movie but I really loved Martin in THe Spanish Prisoner, playing a totally dramatic part.

For me, Planes, Trains and Automobile, followed very closely by The Jerk and Dirty Rotten Scoundrels.

Dirty Rotten Scoundrels was hilarious. The Jerk comes in a close second.
