Antonio's usual thorough list. I can only add one: Professor Dave Jennings (Donald Sutherland) from National Lampoon's Animal House: "I'm expecting papers from some of you on this... Listen, I'm not joking! This is my job!" (one of @ 250 LOL lines from that classic movie.)
John Keating is a particular favorite. That scene at the end, with the desks...that's lump-in-the-throat material for me, every time.

Two more babes for the road: Laura Klein (Isabella Rossellini) in FEARLESS and Jane Spofford (Susan Sarandon) in THE WITCHES OF EASTWICK. Not the traditional teachers (as they taught ballet and music appreciation), but effective nevertheless in two powerful movies centering around spirituality.
