I agree with everyone who said John Goodman in The Big Lebowski and George C. Scott in Dr. Strangelove. 2 of the funniest performances ever, in my opinion.
Also:
-Ben Stiller in The Royal Tenenbaums (some people call Gene Hackman a supporting role, and if I did too, I would definitely put that here. But Hackman was lead, my friends.)
-John Turturro in The Big Lebowski (his 2 minutes of screen time are probably the 2 funniest minutes in the movie, and feature the best line in the movie)
-Marty Feldman in Young Frankenstein (for obvious reasons)
-Joe Pesci in Goodfellas (not only is it a hilarious performance, but it's also an extremely realistic one. That Best Supporting Actor was well-deserved. IMHO, one of the 5 best performances of the 90's. He made it look so easy.)

Dennis Farina in Get Shorty was priceless.
So was John Turturro in The Big Lebowski, and basically anyone in Jay & Silent Bob Strike Back, especially Chris Rock, who cracked me up good there for some reason.

My favorite is Jean Hagen in Singin' in the Rain. She stole every one of her scenes. 'whaddya think i yam...dumb or somethin'?'
Another great one is Peter Ustinov in Spartacus...much needed comic relief. An honorable mention should also go to Daniel Day Louis in A Room With a View-the man you LOVE to hate! "Stop, you look like a Leonardo..."

Maggie Smith and Michael Caine were absolutely HILARIOUS! I don't remember the last time I laughed so hard while watching a movie. The other storylines are boring, but theirs is hilarious! They'd be my choices! Smith won a well-deserved Oscar for her comedic performance. Caine should've at least been nominated.

Wow can't believe im the first one to say this Jeff Daniels in Dumb and Dumber.
I don't consider Jeff Daniels to be a supporting actor in Dumb and Dumber. A movie can have more than 1 lead and I think Daniels is as much a lead as Jim Carey was in that movie.
You listed Vince Vaughn in Old School, well personally, I would list Will Ferrell instead of Vaughn. However, I do think Vaughn should be listed for his utterly hilarious performance in Wedding Crashers along with Isla Fisher.
And Dustin Hoffman in Meet the Fockers.
Mark Wahlberg in I Heart Huckabees. Probably Jude Law as well.
