There are two that I really enjoyed. I really liked "Spaceballs" the spoof on "Star Wars" and I also liked "Galaxy Quest" which spoofed "Star Trek".
So...what is the most entertaining Sci-fi Spoof?

I can't even think of any others.
I enjoyed both Spaceballs and Galaxy Quest for what they were. They weren't "laugh my ass off" comedies but they were entertaining little flicks that generated a decent number of laughs at their intended targets.

Galaxy Quest. I see that I am not alone. Locally (San Francisco) critics did not give it a particulaly worthy review and when I decided to see it it was mainly because of an actor and actress who were in it. Boy was I surprised. I had such a good time that I decided to go again since my reaction was so different. Since then I have rented it and pushed friends to see it. It stands up well for me.

Yes is Spaceballs, even when Galaxy Quest and Mars attack got some hillarious moments.
